(a) Levels of I-κB and a reference protein glyceraldehyde-3-phosphate dehydrogenase (GAPDH) in MCF-7CC, MCF-7TXT8, MCF-7TXT10, and MCF-7TAX-1 cells in the absence or presence of 10 ng/ml Tumor-necrosis factor (TNF)-α. Protein levels were assessed in immunoblotting experiments with quantification of band intensities by using densitometry. The effect of selection for survival in increasing doses of docetaxel on the activity of the NF-κB p65 (b) and p50 (c) subunits in the absence or presence of docetaxel also was examined by using an ELISA. Mean (± standard error) values are plotted, and the significance of differences between MCF-7CC and taxane-resistant cells or differences between treated and untreated cells was assessed by using a Student t test; P values of < 0.05, < 0.01, and < 0.001 are represented by *, **, and *** symbols, respectively.
