Analysis of sodium iodide symporter (NIS) expression. (a) NIS expression assayed with Western blotting in Ad5AMUCH_RSV-NIS-infected cells. Membrane proteins were prepared 24 hours after infection. (b) Radioiodine uptake dose-response. T47D cells were infected at the indicated multiplicity of infection (MOI). Radioiodine uptake was measured 24 hours after infection. (c) Kinetics of NIS-mediated radioiodine uptake. T47D cells were infected at MOI 20 with the Ad5AMUCH_RSV-NIS conditionally replicating adenovirus (CRAd), and 125I uptake was measured daily.
