The YY1.2 mutation disrupts binding of YY1. Electrophoretic mobility shift assay was performed with a probe containing the ying yang 1 (YY1).2 sequence. Nuclear extract from MCF7 and HS578T cells (25 μg) was incubated with either a wild type probe or a mutant probe bearing the YY1.2 mutation. For super-shift experiments, control antibody (Ab; against Ac-H3K9) or an antibody to YY1 was added to the indicated sample. The super-shifted YY1-DNA complex is indicated with an asterisk.
